Active Vitamin D3 Protects Against Diabetic Kidney Disease by Regulating the JNK Signaling Pathway in Rats

چکیده
Diabetic kidney disease (DKD) is an inflammatory caused by metabolic disorder. As important signaling pathway in the response, JNK plays crucial role injury DKD. Vitamin D3 can reduce reaction and delay or even reverse DKD progression. Unfortunately, mechanism which vitamin regulates pathogenesis unclear. This research established a rat model irbesartan were used as interventions. Then, urine blood biochemistry; cytokine (IL-1 IL-6), phosphorylated protein (MEK-4 JNK1/2/3) downstream factor (AP-1 ATF-2) expression assessed. We found that group showed body weight insulin secretion significantly decreased; increased FPG, HOMA-IR lipids; 24-h urinary (UPro) compared with normal group. Additionally, levels of IL-1 IL-6 proteins elevated. These changes improved D3, especially at low dosage. results suggest active protects against rats reducing release, downregulating pathway, inhibiting transcription AP-1- ATF-2-mediated damage. provides new theoretical support for treatment diabetic nephropathy.

JunD protects against chronic kidney disease by regulating paracrine mitogens.
The AP-1 transcription factor, composed of Jun and Fos proteins, plays a crucial role in the fine tuning of cell proliferation. We showed previously that AP-1 complexes are activated during the proliferative response that parallels the development of renal lesions after nephron reduction, but little is known about the specific role of individual Jun/Fos components in the deterioration process. ...
متن کاملVitamin D receptor signaling in podocytes protects against diabetic nephropathy.
Vitamin D and its analogs have antiproteinuric activity and podocytes express the vitamin D receptor, but whether vitamin D signaling in podocytes accounts for this renoprotection is unknown. To investigate this question, we used the 2.5 kb podocin promoter to target Flag-tagged human vitamin D receptor (hVDR) to podocytes in DBA/2J mice. After the induction of diabetes with streptozotocin, tra...
متن کاملEmodin protects against diabetic cardiomyopathy by regulating the AKT/GSK-3β signaling pathway in the rat model.
Diabetes mellitus (DM) has been recognized as a major health problem. Emodin (Emo) has been reported to exhibit protective effects against diabetic nephropathy. However, little has been known about the effect of Emo on diabetic cardiomyopathy (DCM). A type 2 DM model was induced in rats by low dose streptozotocin (STZ) combined with high energy intake. We found that Emo-treated groups displayed...
متن کاملGallic acid protects the liver in rats against injuries induced by transient ischemia-reperfusion through regulating microRNAs expressions
Objective(s): Gallic acid (GA) is a highly effective antioxidant, which its beneficial effects are well known, but its impact on expression of microRNAs (miRs) following hepatic ischemia-reperfusion (I/R) is not well recognized. Therefore, the current research was designed to specify the beneficial effect of GA on miRs (122 and 34a), liver functional tests, and histopathological alterations bey...
متن کاملHydrogen sulfide protects coriander seedlings against copper stress by regulating the ascorbate-glutathione cycle, in leaves
Heavy metals are the cause of major abiotic stresses in plants and a principal contributor to environmental pollution in recent decades. This study investigated the effects of exogenous hydrogen sulfide on the ascorbate-glutathione cycle in the leaves of coriander seedlings under copper stress. Results showed that copper stress not only reduced APX and GR activities but also decreas...
